Acid-induced promoter(PrcfB)

The rcfB promoter, a member of the CRP-FNR positive regulation family, was amplified from chromosomal DNA lactis subsp. lactis IL1403. It is an important part of the P170 promoter regulated by acid. When the external pH value declines to 5.5, rcfB promoter will be highly upregulated.

Results

The A11Sg-AHPM protein was carried out in the lactococcus lactis MG1363 harboring the pMG36e-rcfB-O1/O2-SPusp45-A11Sg-AHPM vector, which was grown in the GM17 broth under different pH. In the 12%SDS-PAGE, one band of size 64 kDa corresponding to the recombinant modified protein was detected at the pH=6.
