Uric acid-induced Uricase generator
This device could produce uricase only when there is uric acid. In the device, HuR is a repressor than can bind to HuO operator to inhibit the gene expression down stream HuO. In the presence of uric acid, the structure of HuR is altered and is no longer able to bind HuO, leading to the down stream gene expression. We inserted a uricase gene (URI) downstram HuO. In this case, uricase is expressed only when there is significant level of uric acid. To better visualize the gene expression, we include RFP(E1010) downstream HuR and a GFP(E0040) downstream URI. In our project, we express it in E. coli. It is hoped that the uricase-expressing E.coli can lower the uric acid concentration in human alimentary canal, so as to prevent gout disease.
