Design Notes

This composite part was designed such that it will express the fusion protein we designed, csgA-CBD. The promoter is a constitutive T7 promoter which requires T7 RNA polymerase to be expressed. The coding region is the curlin precursor protein from E.Coli csgA linked to the chitin binding domain of Bacillus circulans, with a 6x polyhistidine tag on the end. As csgA has well documented amyloid forming behavior we theorized that the fusion protein with a chitin binding domain would make an exceptional biological adhesive for use on a mycelium substrate.
