RBS-ldhD-RBS-lldP

ldhD can produce lactate, and lldP can release it from cytoplasm to the environment.

Characterization

This is a two section for lactate produce and transfer part.

DNA Gel Electrophoretic

After we link ldhD and lldP successfully, we design several tags and then add these tags through PCR with an intention to confirm the expression of ldhD and lldP. As the figure shows, we get the genetic stripe about 3000bp which means the PCR is successful.

As the figure shows, with the help of light and PSI, Synechocystis sp. PCC 6803(cyanobacteria) with ldhD-lldP absorb CO2 into Calvin cycle and produce pyruvate. And then ldhD can transfer pyruvate into lactate which will be transported to the environment by lldP.
