Degradation Part (ptet+Lip+LacI+plac+phi174E)

This part is the best of the many degradation circuits we have built. It is made up of Lip and suicide switch.We cocultured

BBa_K2685007 and BBa_K2685016 at different concentrations of tetracycline. Lip degradation effect was measured by

using unit fluorescence intensity because Lip degraded tetracycline caused BBa_K2685007 concentration growth thereby

increased its unit production fluorescence intensity. From the pictures we can see that the unit fluorescence intensity

of BBa_K2685007 mixed with BBa_K2685016 is generally higher than that of BBa_K2685007 alone.This further indicates that

Lip can efficiently degrade tetracycline in the environment.

The figure below shows the common validation of the different degradation circuits we built. Compared with the MNP and

bi-enzyme degradation circuits, BBa_K2685016 is one of the parts with the best degradation effect.
