OmrB

OmrB is a small RNA that takes part in regulating the outer membrane protein composition. Research has shown OmrB to negatively regulate the expression of membrane proteins such as ompT and cirA. The effect of OmrB sRNA on ompT and cirA expression depends upon RNAse E enzyme and involves direct base pairing at the 5’ end of OmrB. The action of OmrB is also Hfq dependant. It is the 5’ end of OmrB that is involved in the sRNA’s regulatory function. A deletion mutant would result in a change in outer membrane protein composition, whilst overexpression of OmrB causes downregulation of CsgD expression. This results in inhibiting the formation of bacterial curli.

Usage and Biology

This synthetic version of OmrB follows the same principles as its wild type partner to regulate gene expression. This modified construct, however, can target any gene of interest due to its sequence which contains a BaeI restriction enzyme site. After digestion with this restriction enzyme and ligation with a target sequence, OmrB can regulate the expression of the gene that contains the complementary regions for its targeting sequence. Ligation with the desired targeting sequence does not create any scar in the sRNA sequence.
