{R6K} conditional origin of replication in BBb

The R6K origin of replication requires a protein called pi encoded by the pir gene for function. At higher expression of pir in strains containing the pir116 allele, R6K plasmids replicate at high copy comparable to pUC plasmids. At lower expression levels such as observed in pir+ strains, they express at medium copy comparable to p15A plasmids. For most laboratory E. coli strains, there is no pir gene, and no replication is observed. In house, this part is referred to as pBca9145-Bca1182 {R6k}.
