The final RFP/OD values for this riboswitch are shown as "#3" in the graph below. Ideally, the riboswitch would show high RFP/OD with xanthine and not the other conditions. However, the data suggest that the riboswitch actually remained strongly theophylline specific.

To compare the theophylline induction of this riboswitch compared to the original RS-C, the fold-induction of each theophylline-specific riboswitch was calculated by dividing RFP/OD in the presence of theophylline by the RFP/OD without any ligand present. The chart below shows the fold-induction of wildtype (WT) RS-C and RS-C mutants #3, 7, 8 over a 10-hour experiment.

RS-C mutant #3 has much higher induction than the WT RS-C or RS-C mutants.
