EL-222 is a light-sensitive transcription factor that binds to DNA and drives transcription under blue light illumination. EL-222 consists of a LOV-HTH (helix-turn-helix) domain and VP16 activating unit. The LOV-HTH allows for light-gating of DNA binding; the LOV portion of the protein covers the HTH portion of the protein that binds DNA - under blue light exposure, the LOV domain changes conformation and reveals the HTH DNA binding site. The HTH binds to a sequence of c120 repeats. Once bound, the VP16 activating unit promotes transcription factors in the cell to drive genes downstream of the c120 repeats.
