Temperature-sensitive repressible system

In this construct, the temperature sensitive TlpA36 protein will be produced with the addition of a upstream promoter (as specified by the user), and depending on the temperatures, it will either form Tlpa36 dimers that are capable of repressing the pTlpA promoter (in temperatures below 36 degrees) or stay in monomeric form (above 36 degrees) which makes the protein unable to repress the promoter.

As a proof of concept that the temperature system is fully functional, we had constructed and characterised an ITPG-inducible temperature system (BBa_K2447014) and coupled it to a downstream GFP reporter.
