DtxR inverter using TetR

This part inverts the signal produced by the interaction between DtxR protein and the cognate operator.

Usage and Biology

Wen Fe2+ is added to a system where the DtxR protein is present (which can be achieved by the use of our dtxR generator) the protein enters in it's active state and is able to repress gene expression through its cognate operator. In our inverter system we have a modified constitutive promoter (BBa_X) that produces TetR in the absence of active DtxR. The produced TetR in turn can repress a second promoter with the tetR operator. In the presence of Fe2+ the tetR expression would cease and permit reporter production.
