mEmerald

mEmerald is a derivative of the Green Fluorescent Protein from Aequorea victoria with improved features in terms of intrinsic brightness, photostability, resistance to bleaching, maturation and folding at 37°C [1]. The protein contains six mutations compared to wild-type: F64L; S65T; S72A; N149K; M153T; I167T; A206K [2].

This protein has been specifically optimized for Escherichia coli K12 with help of JCAT ([http://www.jcat.de/]). Before optimization, the CAI-Value of the original sequence was 0.419 with a 62% GC-Content. The optimization allows a CAI-Value for the improved sequence of 1.0 with a 48% GC-Content.

Usage and Biology

Applications are various: Fusions, promoter & secretion studies, FRET and other quantitative imaging techniques.

Our team used this codon optimized mCherry to build the Universal Biosensing Chassis (BBa_K2448023).
