This part was constructed by combining BBa_K2488000 and BBa_ K1321340.

Here you can occur the compatibility of phytase and cellulose binding domain.

To characterize this part we introduced it on pUV3 to yeast yarrowia lipolytica.

In comparison with an ordinary phytase inserted to the pUV3, phytase combined with cellulose binding domain is resistant to yeast enzymes thanks to domain’s compact structure. So it easily can get to the gut, where effectively breaks down the phytin.

Thus, dCBD acts not only as super compact protein domain but as a barrier between phytase and destructive enzymes of yeast cells.

Thanks to this improvement the domain acquires a number of new advantages.The first thing is that now this domain can be used for two-step purification on an affinity chromatography.Moreover, being a part of new biobrick and interacting with phytase, cellulose binding domain acquires such a new advantage,as viability in the acidic pH environment of the intestine(pH level=5-7)

We confirmed that a new improved part worked accurately.
