Design Notes
The P32 promoter and RBS sequence are natively found in the genome of Lactococcus lactis. However, we ordered the promoter and RBS sequence as a gBlock using the sequence information provided by Zhu et al. (2015) in "Isolation of strong constitutive promoters from Lactococcus lactis subsp. Lactis N8" (https://doi.org/10.1093/femsle/fnv107). Our lab uses the Lee/Dueber YTK Golden Gate standard, which, in terms of part overhangs, is different than the iGEM PhytoBrick standard. Thus, we had to redesign our P8 promoter sequence to contain the proper overhangs specified by the iGEM PhytoBrick standard.
