RafD, an invertase followed by HlyA, a secretion signal
It is the coding sequence of RafD followed by a secretion signal HlyA. And a GGGGS linker is between them. RafD is a invertase which can catalyze the hydrolysis of sucrose to glucose and fructose so that the bacterial can use sucrose in the medium. HlyA tag can make it possible for RafD to be secreted outside the cell, which can make not only the producer use sucrose, but also other bacteria which can not synthesize this protein. GGGGS linker is added between RafD coding sequence and HlyA signal peptide so that adding of HlyA will not influence the folding and function of RafD. The secretion of RafD also needs help of a secretion system, which can be seen in the main page of part BBa_K1166002.
