Applications of BBa_K1900003

This part works to supplant the antibiotic efflux function of a deletion strain of E. coli that lacks the gene. The deletion strain shows little resistance to novobiocin and erythromycin but is able to resist both of these when transformed with the tolc gene. Below a comparison is made between the strain lacking the tolc gene, and the strain with the E. coli tolc gene. A Kirby Bauer assay was done to assess efflux function. The larger the zone of inhibition, the less resistant the bacteria is to the antibiotic. Additionally, the E. coli tolc gene was compared to other tolc genes from various gram negative bacteria. These include Serratia marcescens and Klebsiella pneumonia. A 3D model of the protein is also shown below.
