Because we did not observe maximum GFP production for arsenate concentrations under 15 mM, we performed a dose response study with arsenate concentrations ranging from 0 mM to 30 mM. We grew cells in a total volume of 1.5mL LB in 37°C for 24 hours in a plate with a breathable cover, starting with an OD600 of 0.01. We performed this experiment three times with three replicates per experiment. We included wild type JM109 cells as a negative control and pColi JM109 in 100 uM rhamnose as a positive control for GFP production. (pColi is a plasmid that is known to produce GFP when induced with 100uM rhamnose.) We concluded that significant levels of GFP are produced by JM109 cells transformed with our plasmid at KH2AsO4 concentrations of 15 mM and above.
