We transformed our plasmid containing the arsR promoter and arsR gene, amp resistance, and GFP into JM109 E. Coli cells. Wildtype JM109 wild type cells have been reported to have arsenic resistance up to 20 mM (Edmundson, M.C., Horsfall, L. 2015. Front Bioeng Biotechnol. 3(160): 1-11.) We grew these cells in varying potassium arsenate concentrations in LB between 0 mM and 30 mM. Cells were grown overnight at 37ºC and GFP levels were measured 24 hours later. Relative GFP was calculated using Relative GFP = (GFP of sample - GFP of blank) / (OD600 of sample - OD600 of blank).
