For the 2016 Team Rice project, we built a biosensor construct in E. coli with fdhF and iRFP670 to fluoresce in the absence of oxygen. Specifically, the cofactor fdhA binds to formate, a metabolite in anaerobic respiration, to upregulate transcription downstream of fdhF. However, despite this description from literature, our assay results indicate that transcription was downregulated in anaerobic conditions.
