We have linked this circuit in pSB1K3, pSB3K3 and pSB4K5 and tested them in E.coli BMTop10. We cultured our bacteria overnight and then tested fluorescence intensity to indicate the expression level of circuit in different plasmids.
The fluorescence intensity is shown in Fig.1. As,we all know,pSB1K3 has the highest copy number and the copy number of pSB3K3 is little higher than the copy number of pSB4K5. And according to the result, the fluorescence intensity of bacteria containing pSB4K5 is higher than containing pSB3K3. The fluorescence intensity of bacteria containing pSB1K3 is the lowest. It means our system is working. Our killer would express more with less plasmids.
