Synthetic Enhancer Project: 2X TetO Binding Cassette (55AS)

55AS binding cassette has two TetO binding sites. Based on the amount of aTc in the system, which binds and inactivates TetR repressor, the amount of Tetr bound to these two TetO varies and thus flexibility of the DNA is also changing and ultimately the looping between the enhancer and promoter and output levels. These two TetO sites on 55AS allows for three discrete states: unrepressed, intermediate, completely repressed. This part is from a plasmid gifted by Amit et. al 2011.
