Strong Anderson / Taz

BBa_K1909010 is a construct of strong Anderson promoter BBa_K081005, a strong ribosome binding site and the receptor tar-envZ, part BBa_C0082, which is a combination of the chemotaxis receptor Taz capable of sensing aspartate, and the cytosolic domain of osmosensor kinase EnvZ.

Functional Parameters

This combination has the highest expression of the three combinations BBa_K1909010, BBa_K1909011 and BBa_K1909012. Testing the combination showed that the cells grew very slowly (for miniprep culture more than 48 hours). We assume that this combination (strong promoter and strong RBS) induces production of toxic amounts of the transmembrane receptor.
