eGFP

Modification

In iGEM 2016, our team, SYSU-MEDICINE, had constructed a new composite part BBa_K1993008 in order to improve the positioning function of this previous part. Our new composite part contains gene Luciferase, IRES and eGFP, which help users observe biological processes in vivo and in vitro.

the fluorescent protein eGFP, codon usage of E. coli, works also in Phaeodactylum tricornutum. The excitation peak is at 488nm and the emission at 507nm.

Usage and Biology

In the following picture we demostrate that both promotors (i.e. PNR (496) and PfcpB (416)) work properly as shown by the expression of eGFP (green, second panel). The red color (third panel) is due to autoflourescence of the plastides and indicates their localisation within P. tricornutum. The very left and the very right panel represent DIC and an overlay of green and red, respectively.
