Design Notes

Design of the PyeaR-serA BioBrick was accomplished by combining our own serA BioBrick with the yeaR promoter and RBS used previously. Due to the inability to obtain just the yeaR promoter and RBS, a fragment that contained the yeaR promoter and RBS was synthesized by IDT with the BioBrick Prefix and suffix and then ligated to our own BioBrick. The terminator was from the part BBa_B0015 found in the distribution kit. All three parts were ligated together using 3A assembly. The terminator and serA were ligated first, and once that was completed the promoter and RBS were attached.
