This His tag was codon optimized for E.Coli and specific for the EP-B2 protein. It's a marker for the EP-B2 protein that can be used for identification. Another purpose of the His tag is protein purification, specifically The EP-B2 protein. When planning our experiments, we were going to do a Western Blot, however, we ended up not needing to do a Western Blot, because our other assays provided sufficient results. Since we did not do a Western Blot the His tag was not actually a necessary part of the construct. If we end up doing follow-up experiments this His tag will be necessary.
