Design Notes

The length of DNA BMs and intervening sequences is designed based on the typical in vivo B-type DNA structure, and the Structural Basis for Sequence-Specific Recognition of DNA by TAL Effectors.

This scaffold is designed to avoid steric hindrance and to ensure the spatial orientation of enzymes. A 6-basis intervening sequence was added between BMs.
