slrR with strong promoter and RBS

SlrR is an important transcriptional regulator in Bacillus subtilis. SlrR represses the sigma(D)-dependent flagellar genes and activates the eps and yqxM operons. Therefore it is has a controlling factor in biofilm formation. By overexpressing slrR, with the use of a strong promoter (BBa_K090504) and a strong RBS (BBa_K780001)(BBa_K780001), a robust biofilm can be formed. slrR with the promter and RBS was integrated via BBa_K823023 and via BBa_K1597001.

slrR mutant with an overexpression of slrR shows a bigger biofilm than the wildtype B. subtilis 3610 ΔcomI control. This phenotype could be due to the overexpression of the eps operon, leading to many extracellular polysaccharide substances. Resulting in a larger biofilm. The combination of the abrB knockout and slrR+ resulted in a biofilm size to the control. The process behind the this interaction is not known, however comparing these phenotypes clearly shows an interaction.
