slrR with strong promoter and RBS

SlrR is an important transcriptional regulator in Bacillus subtilis. SlrR represses the sigma(D)-dependent flagellar genes and activates the eps and yqxM operons. Therefore it is has a controlling factor in biofilm formation. By overexpressing SlrR, with the use of a strong promoter (BBa_K090504) and a strong RBS (BBa_K780001), a robust biofilm can be formed. slrR with the promter and RBS was integrated via K823023 and via K1597001

slrR mutant with an overexpression of slrR has shows a bigger biofilm than the B. Subtilis comI control. This phenotype could be due to the overexpression of the eps operon, leading to many extracellular polysaccharide substances. Resulting in a larger biofilm. The combination of the abrB knockout and slrR resulted in a comparable biofilm to the control. This suggests that an abrB knockout together with slrR affect each other. The process behind the this interaction is not known, however comparing these phenotypes clearly shows an interaction.
