Design Notes

We used the promotor sequence from "Genomic Mining of Prokaryotic Repressors for Orthogonal Logic Gates" Stanton et al http://www.nature.com/nchembio/journal/v10/n2/full/nchembio.1411.html

Stanton et al., (2014) overlapped the operator sequence for the SrpR repressor over the -35 site of BBa_J23119, a strong, constitutive Anderson family promoter, meaning when the repressor binds to its operator sequence sigma factor cannot recognise the promoter (and therefore attract RNA polymerase) and transcription cannot start.
