Glycoprotein G epitopes derived from Herpes Simplex Virus-1

This part contains the sequence of an immunogenic part of glycoprotein G from Herpes Simplex Virus-1 (HSV-1). It was obtained from Liu et al. (2005) who generated the sequence for the development of improved diagnostic applications. In 2013, the same group made use of its specific antibody binding properties to develop a protein microarray based diagnostic tool that allows distinguishing between several Herpes Viruses.

The part is composed of amino acids 52 to 181 of glycoprotein G and contains several immunogenic epitopes. In general, it is used for determination of the infection status of immunosuppressed patients, yound children and pregnant women with HSV-1. The most crucial point in the diagnosis of HSV-1 is to distinguish it from HSV-2. Those two types of Herpes Viruses are so closely related to each other that they share their immunogenic sequences. Ashley et al. (1998) showed that this is not true for glycoprotein G. Accordingly, commercial kits based on the detection of antibodies have been developed with aid of glycoprotein G.

The iGEM standard backbone pSB1C3 was used for shipping the part to the registry. Cloning of the part into this backbone was performed by Gibson Assembly and the sequence was verified afterwards.
