Predator Molecule Generator Sensing Test (GFP output)

Usage:

  • This part was created by iGEM@Imperial 2006 to be used as the predator sensing test construct in their 2006 project [http://openwetware.org/wiki/IGEM:IMPERIAL/2006 'Engineering a Molecular Predation Oscillator'] to characterize the sensitivity of the predator cell towards the prey moleculte Acyl-Homoserine Lactone (a quorum sensing molecule).

Biology:

  • In the presence of AHL, AHL and LuxR form a heterodimer, which activates the promoter luxpR and thus expression of the genes downstream of the promoter. Depending on the levels of AHL, the level of expression of LuxR and GFP vary.
  • The behaviour of this part was successfully modelled, generating a [http://openwetware.org/wiki/IGEM:IMPERIAL/2006/project/Oscillator/project_browser/Test_Sensing_Predator_Construct/Modelling transfer curve].
  • The part was successfully tested using this [http://openwetware.org/wiki/IGEM:IMPERIAL/2006/Protocols/T9002 testing protocol] and [http://openwetware.org/wiki/IGEM:Imperial/Results/J37016 experimental data] from testing was documented.
