Design Notes

The design is based on sequences and methods proposed in Elowitz et. al. 2007 The sequences with the desired operator sites where chosen from the sequences in the supplementary data, and put together in silico. The Promoters where than ordered as two primers with a 20 base pair overlap so that we could use it for primer extension. The protocol for primer extension can be found [http://openwetware.org/wiki/Knight:Annealing_and_primer_extension_with_Klenow_polymerase here]

Source

The sequences on which this promoter is based can be found in Elowitz et. al. 2007 in the supplementary data.
