Usage and Biology

The 2013 iGEM Freiburg 2013 biobricked an inactive Cas9 (dCas9), which binds to DNA but cannot cleave it anymore and fuctions as a DNA binding protein [X]. We improved this enzyme by a single nucleotide mutation to a nickase. The Cas9-Nickase is capable to induce single strand breaks in the genome, thus leading to a simple method of gene editing [Y].
