Omega undercover

The omega undercover vector has a BioBrick backbone but is compatible with GoldenBraid 2.0, a Golden Gate-based assembly system. It contains two BsmBI restriction sites which leave the required overhangs to perform a GoldenBraid ligation of two transcriptional units in alpha vectors. We call it "omega undercover" as it works in a similar way of the GoldenBraid 2.0 omega vector but with a pSB1A3 backbone.


Usage and Biology

The omega undercovered rises as a way of trying to make BioBricks compatible with other Golden Gate-based assembly systems. It contains two BsmBI restiction sites which after digestion leave the overhangs required to perform a ligation of two transcriptional units which follow the required grammar. This grammar consists of the sequence GGAG on the 5' end and CGCT on the 3' end. Our team successfully used this vector in order to send Part:BBa_K1554004 and Part:BBa_K1554005 to the Registry of Parts. The molecular process involved in such ligation is explained in the following figure.
