plac+taRNA+ter
The Plac 0-1 promoter is a hybrid regulatory region consisting of the promoter P(L) of phage lambda with the cI binding sites replaced with lacO1. This regulatory region will be repressed by LacI or induced by IPTG. taRNA is taR12 which is bioBricked version of Isaacs' riboregulator trans-activating key. After the PLlac 0-1 promoter being activated, the transcription of taR12 key will increase and the increasing taR12 key will activate the corresponding crR12 lock. The hybrid design which contains two different control systems allows for the strong and tight control of the expression of downstream gene.
