Results of a highly-efficient one-pot one-step GoldenGate cloning reaction in which a Golden Gate promoter module was inserted into BBa_K1467100 (“Pro Flipper”), replacing the RFP reporter. The BsaI recognition sites have been eliminated by the reaction, converting the part to a standard BioBrick. The very low number of red (RFP-expressing) colonies indicate the extremely high-efficiency of the reaction.
