No specific design considerations other than ensuring that the sequence was in reverse.

Made by PCR using E1010 as a template and primers XXXX and YYYYY to incorporate prefix at the 3' end of the rfp gene and the suffix 5' end of rfp. The final 'C' of the bar code was omitted, with no effect on rfp function.
