RFP coding device - Golden Gate Module Flipper
The colonies are clearly red in color under natural light after about 18 hours. Smaller colonies are visibly red under UV. The RFP part does not contain a degradation tag and the RBS is strong. LacI sensitive CAP sensitive This part is a variation of Part:BBa_J04450 with additional flanking sequences recognised by the enzyme BsaI that allow standard GoldenGate Modular Cloning (MoClo) Promoter+5' untranslated leader (Pro+5U) modules to be 'flipped' in a one-pot, one-step, digestion-ligation reaction with BsaI, thus converting the MoClo part to a BioBrick part. When cloning is successful the colonies become white as the RFP sequence is replaced by the new part.
