PpelA

PpelA is an inducible pectate lyase promoter isolated from Aspergillus nidulans. It is repressed by glucose and inducible by polygalacturonic acid. Thus, controlled homologous and heterologous gene expression can be achieved by cloning the desired gene downstream of PpelA and adding glucose or polygalacturonic acid to the fungal culture. PpelA was constructed by digesting the fungal transformation vector pHNU3pelA with BamHI and KpnI restriction enzymes and subsequently ligating the digested fragment into a pSB1C3 backbone appended with BamHI and KpnI restriction sites.
