Terminator BBa_B0015 with inversed Pre- and Suffix- DarR ORF with inversed Pre- and Suffix

  • This composite part is composed of BBa_K1045001, an inverse DarR coding sequence, fused to an inverse terminator, BBa_K1045009. DarR is a transcriptional repressor binding to the DarR operator sequence. Repression by DarR is enhanced by c-di-AMP. In E. coli, DarR seemed to act as a strong repressor even without c-di-AMP ([http://2013.igem.org/Team:Goettingen iGEM Göttingen 2013]). BBa_K1045016 could be cloned downstream of inverse promoter-RBS constructs (compare BBa_K1045010 (inverse RBS) and BBa_K1045011 (inverse promoter)) allowing it to be expressed (for example, see BBa_K1045017).
