Inducible ampicillin resistance cassette (regulated by LasR + N-3-oxododecanoyl-HSL)

This device is a combination of BBa_K1073003 and BBa_K1073000 and mediates an inducible ampicillin resistance. It includes the inducible promoter of the las quorum sensing system of Pseudomonas aeruginosa followed by a RBS and the ampicillin resistance gene coding for beta-lactamase.

Usage and Biology

The promoter is activated in the presence of LasR in combination with the autoinducer molecule N-3-oxododecanoyl-homoserine lactone produced by LasI. The expression of the beta-lactamase can therefore be induced and tuned by supplying LasR and N-3-oxododecanoyl-HSL.

Note that the promoter is leaking to a certain degree when not activated. For further information and experience on this device see BBa_K1073034.
