violacein A coding sequence
Violacein A (Vio)A is the first of 5 protein coding sequences contained in the Violacein operon originally derived from the organism, Chromobacterium violaceum. The entire operon was submitted to the registry by the Cambridge team in 2009 as K274002. Together these five proteins form a metabolic pathway that produces the violet pigment violacein when expressed in E. coli. Like its other four partners we have appended the coding sequence with the ribosome binding site BBa_B003 so that operons that vary in gene order can be created by standard assembly means. Here we use these parts to show gene order affects pigment colour.
Usage and Biology
Features: 1. Derived from the violacein operon constructed and submitted by Cambridge 2009 (BBa_K274002). 2. Upgraded from pSB1K3 to pSB1C3 standard 10. 3. Operon Orfs are now individual parts that can be assembled in different combinations and permutations. 4. Each Orf shares a common and improved rbs for more rapid colour development. 5. All have been characterized and are functional. 6. The VioABCE combination produces a new pigment which we call “Royal Blue” Sequence and Features
