B0034-RiboJ-φR73

For detailed information concerning Band-pass Filter, please visit 2013 Peking iGEM Band-pass Filter

Components and Characterization

K1031011 consists of RBS B0034[1], ribozyme RiboJ and transcription factor φR73 delta. φR73 functions as an activator of hybrid promoter. It is the activation node of the Band-pass Filter circuit (Fig 1).

Fig 1 Activation circuit of Band-pass Filter. The green oval represents RBS B0034. Ribozyme RiboJ is marked in dark blue. φR73 coding sequence is shown as square with dark red. φR73 activates hybrid promoter, which is marked with red and blue, via binding upstream activating sequence. Then the expression of sfGFP is enhanced.

Data shown

Previous work done by Dr. Lou Chunbo showed φR73 had obvious activating effect to downstream promoter. Molecular cloning which connected φR73 with Psal promoter enables the circuit to response to salicylic acid (Fig 2).
