DarR reporter system
This composite part encodes a reporter system to monitor the repression mediated by DarR. DarR is a transcriptional repressor that binds to the DarR operator sequence blocking transcription of the downstream gene. These effects are enhanced by c-di-AMP. In part BBa_K1045017, the DarR operator is placed in between a strong promoter BBa_J23110 and a GFP generator BBa_E0240. DarR itself is part of an inverted expression unit with a weak promoter BBa_J23117. Transcription of the DarR mRNA might be terminated by an inverse terminator BBa_K104509. This system was designed to screen for compounds similar to c-di-AMP.
