Design Notes

We had designed the entire protein sequence from scratch, repeating the Threonine-Lysine amino acid pair a total number of 216 times. We then randomised the codon usage for optimization in E.coli.

This protein is one of our own design. Created for use in an MRI to provide CEST contrast. Whilst we designed the sequence ourselves the idea for using Poly amide amino acids came from the following article: "New “Multicolor” Polypeptide Diamagnetic Chemical Exchange Saturation Transfer (DIACEST) Contrast Agents for MRI" by McMahon et al. Here they outline the use of Poly amide amino acids chains, although the chains used here have a length of 12 amino acids. To increase the amide amino acid concentration we therefore decided to increase this to 432.
