Every attempt to culture in broth has failed. This part grows on plates, but the assumption that either the promoter is leaky enough or that light is circumventing our barriers. Perhaps if colony selection and culturing occurs in an environment with filtered/screened light, we would be more successful; however our facilities are ill equipped to explore this option.
