Applications of BBa_K732000

E. coli were transformed with pET30a vector containing His-tagged hCG-β under the control of a strong promoter and the lac operator. In the basal state, lacI repressor binds to the operator and blocks transcription of hCG-β. Cells were induced with IPTG, which causes a conformational change in the repressor that unblocks transcription.

Folded protein mixture was applied to a pregnancy test kit, and the control band appeared. The test strip was allowed to sit overnight, and by the next day the test band had appeared, demonstrating detection of hCG-β.

Test band was not observed when extract was applied without folding solution.
