PAGFP
PAGFP is a photoactivatable green fluorescent protein. PAGFP remains unactivated when exposed to 450-550nm light but fluoresces when exposed to a wavelength of 400 nm. It is used a reporter tag; by expressing the protein in cells or localizing to sub-cellular locations, one can gain temporal and spatial information within the cell by tracking organelles, proteins, and the whole cell itself. One can visualize the PAGFP with the same filter sets used for other green fluorescent proteins.
In these images, PAGFP is targeted to the peroxisome. The left image is prior to photoactivation and the right image is post activation.
