The plasmid seems to be missing the X and S restriction sites. Digestion by these enzymes fails GelK206009.jpg

Agarose electrophoresis of part K206009 as ordered from the registry in spring 2011 digested with several iGEM enzymes. nd stands for "non digested"

As shown above, the digestion by P linearizes the plasmid as expected but X and S have no effect compared to the non digested plasmid. Moreover, double digestion by X+P only linearizes the plasmid.


This is an intermediate part for the Jammer subproject. The kit plate DNA of this part from Spring 2012 distribution is incorrect. University of Calgary 2012 iGEM team built BBa_B0034 and BBa_K082003 and resubmitted this part as BBa_K902016.
