Design Notes

We think it improved BBa_I13504(RBS-GFP-Terminator) in parts.igem. However, we think the lac promoter cannot work well on pSB1C3 for there is not lacI gene on it. And K12 strains can not express enough lacI to repress the expression of plac. So the leak expression of lac promoter is quite high. We use glucose in our later experiments to repress plac. But the results are not that good.
